코르티코트로핀 방출 호르몬
Corticotropin-releasing hormone코르티코트로핀 방출 호르몬(CRH)(코르티코트로핀 방출 인자(CRF) 또는 코르티콜리베린으로도 알려져 있다; 코르티코트로핀은 스트레스 반응에 관여하는 펩타이드 호르몬이다. 코르티코트로핀 방출 인자 계열에 속하는 방출 호르몬이다. 인간에서는 CRH 유전자에 의해 암호화된다.[5] 그것의 주요 기능은 HPA 축의 일부로서 ACTH의 뇌하수체 합성의 자극이다.
코르티코트로핀 방출 호르몬(CRH)은 196-아미노산 프리프로호르몬에서 유래한 41-아미노산 펩타이드다. CRH는 스트레스에 반응하여 시상하부의 심실핵(PVN)에 의해 분비된다. CRH 생산량 증가는 알츠하이머병 및 주요 우울증과 관련이 있는 것으로 관찰되었으며,[6] 자가 열성 저체온성 코르티코트로핀 결핍은 저혈당증을 포함한 다발적이고 잠재적으로 치명적인 대사 결과를 초래한다.[5]
CRH는 시상하부에서 생성되는 것 외에 T 림프구와 같은 말초 조직에서도 합성되어 태반에서도 고도로 발현된다. 태반에서 CRH는 잉태 기간과 분만 및 분만 시기를 결정하는 마커다. CRH 순환수치의 급격한 증가는 분열을 시작할 때 발생하며, 이는 대사 기능 외에도 CRH가 분열을 유발하는 계기가 될 수 있음을 시사한다.[5]
호르몬 작용
CRH는 시상하부의 대심실 핵 내에 있는 파보세포 신경내분비 세포에 의해 생성되며, 이러한 뉴런의 신경 분비 단자에서 시상하모-하이포피질 포탈 시스템의 1차 모세혈관 플렉서스로의 중간에서 방출된다. 포털 시스템은 뇌하수체의 전엽으로 CRH를 운반하는데, 이때 피티코트로프를 자극해 부전구균 호르몬(ACTH)과 기타 생물학적으로 활성 물질(β-엔돌핀)을 분비한다. ACTH는 코티솔, 글루코코르티코이드, 미네랄코르티코이드, DHEA의 합성을 자극한다.[7]
단기적으로 CRH는 식욕을 억제하고, 주관적인 불안감을 증가시키며, 주의력 고양과 같은 다른 기능을 수행할 수 있다. CRH의 원위 작용은 코티솔의 작용을 통한 면역억제 작용이지만, CRH 자체는 실제로 염증을 증가시킬 수 있는데, 이것은 다발성 경화증 연구에서 조사되고 있는 과정이다.[8]
정신의학
CRH-1 수용체 길항제 펙사이어폰트는 현재 일반화된 불안장애 치료로 조사 중이다.[9] 또 다른 CRH-1 길항제인 안탈라민은 불안, 우울증, 기타 조건의 치료를 위해 동물 연구에서 연구되었지만, 이 화합물을 이용한 인간 실험은 수행되지 않았다.
또한 자살한 사람들의 뇌척수액에서 비정상적으로 높은 수준의 CRH가 발견되었다.[10]
최근의 연구는 CRH1 수용체의 활성화와 알코올 섭취에 수반되는 행복감을 연관시켰다. 화이저가 개발한 CRH1 수용체 길항제 CP-154,526은 알코올 중독 치료 가능성에 대해 조사 중이다.[11][12]
알파-헬리컬 CRH-(9–41)는 CRH 길항제 역할을 한다.[13]
파티셔닝에서의 역할
CRH도 태반에 의해 합성되어 임신 기간을 결정하는 것 같다.[14]
출산 직전 임신 말기에 레벨이 상승하고 현재 이론은 CRH의 세 가지 역할을 제안한다.[15]
- 태아 부신에 대한 작용에 의해 직접, 그리고 산모의 뇌하수체를 통해 간접적으로 디트로피안드로스테론(DHEA)의 수치를 증가시킨다. DHEA는 자궁수축을 준비하고 자극하는 역할을 한다.
- 자궁 내 조직에서 프로스타글란딘의 가용성 증가. 프로스타글란딘은 자궁수축을 활성화시킨다.
- 분리에 앞서 근막내 cAMP 수준을 증가시킴으로써, 역할을 억제할 수 있다.
문화에서, 영양성분 CRH는 프로게스테론에 의해 억제되며, 이것은 임신 기간 내내 높은 상태를 유지한다. 이 프로게스테론 블록을 들어올리기 전에 분리에 앞서 증가하는 글루코코르티코이드와 카테콜아민에 의해 방출이 촉진된다.[16]
구조

비매머리아 척추동물에서의 역할
포유류에서, 연구는 CRH가 유의미한 음낭성 효과를 가지고 있지 않다는 것을 암시한다. 그러나 모든 비인종 척추동물의 대표자리에서 CRH는 피질방성 기능 외에도 갑상선 축을 제어하기 위해 TRH와 작용하는 강력한 음향성 기능을 가지고 있는 것으로 밝혀졌다(TRH는 일부 종에서 CRH보다 덜 강력한 것으로 밝혀졌다).[19][20]
상호작용
코르티코트로핀 방출 호르몬은 그 효과를 유도하기 위해 수용체 코르티코트로핀 방출 호르몬 수용체 1(CRFR1) 및 코르티코트로핀 방출 호르몬 2(CRFR2)와 상호작용하는 것으로 나타났다.[21][22][23] 시상하부(PVN)의 설치성 대뇌실핵에 CRF를 주입하면 CRFR1 발현이 증가할 수 있으며, 표현력이 증가하여 우울증 같은 행동을 유발할 수 있다.[24] CRF와 그것이 상호작용하는 수용체에 관해서도 성 차이가 관찰되었다. CRFR1은 남성에 비해 암핵, 후각결절, 회전근심실핵(AVPV)에 더 높은 수준으로 존재하는 것으로 나타났으며, 남성의 볼륨은 선조체 말단부의 침대핵에서 여성에 비해 CRFR2의 수치가 증가하는 것으로 나타났다.[25]
