라이아노딘 수용체 2
Ryanodine receptor 2라이아노딘 수용체2(Ryanodine receptor2, RYR2)는 라이아노딘 수용체의 한 종류로 주로 심장근육에서 발견되는 단백질이다.인간에서는 RYR2 유전자에 의해 암호화된다.[5][6][7]심장 칼슘 유도 칼슘 배출 과정에서 저장된 칼슘 이온의 사코플라즘 방출은 RYR2가 주요 중재자다.
구조
이 채널은 RYR2 호모테트라머와 FK506 바인딩 단백질로 구성되어 있으며, 1:4 확률적 비율로 발견된다.칼슘 채널 함수는 결합 차이 및 기타 요인에 의해 RYR2 단백질과 상호작용하는 특정 유형의 FK506 이소머에 의해 영향을 받는다.[8]
함수
RYR2 단백질은 systole 동안 심장근육에 이온을 공급하는 사코플라스믹 레티쿨룸에 위치한 칼슘 채널의 주요 성분으로 기능한다.심장근육수축을 가능하게 하기 위해 혈장막의 전압 게이트 L형 칼슘 채널을 통한 칼슘 유입을 통해 칼슘 이온이 사코플라스믹 레티쿨럼에 위치한 RYR2에 결합할 수 있다.이 결합은 RYR2를 통해 사르코플라스믹 레티쿨룸에서 시토솔로 칼슘이 방출되게 하고, 트로포닌의 C영역에 결합하여 트로포닌을 이동시키고 미오신 ATPase가 작용하도록 하여 심장근육수축을 가능하게 한다.[9]RYR2 채널은 심근세포 수축에 대한 역할 외에도 미토콘드리아 대사, 유전자 표현, 세포 생존을 포함한 많은 세포 기능과 연관되어 있다.[10]
임상적 유의성
라이아노딘 수용체 제품군, 특히 RYR2 수용체의 유해한 돌연변이는 급성 및 만성 심부전을 모두 유발하는 병리학적 별자리로 이어지며, 집단적으로 "Ryanopathies"라고 알려져 있다.[11]
RYR2 유전자의 돌연변이는 카테콜아민거 다형성 심실빈맥과 부정맥 유발 우심실 이형성증과 관련이 있다.[12]
최근 아미쉬 커뮤니티의 몇몇 젊은 개인들(그 중 4명은 같은 가족 출신)의 갑작스러운 심장마비로 돌연변이인 RyR2 유전자가 균일하게 복제된 것으로 추적되었다.[13]정상(야생형) RyR2는 주로 심근(심근)에서 기능한다.
유전적으로 감소된 RYR2를 가진 생쥐는 낮은 기저심박동수와 치명적인 아리스티미아를 보인다.[14]
