프로스타글란딘 G/H 싱타아제 1, 프로스타글란딘-엔도페록사이드 싱타아제 1 또는 프로스타글란딘 H2 싱타아제 1로도 알려진 사이클록시제네아제 1(COX-1)은 인간에서 PTGS1 유전자에 의해 인코딩되는 효소다.[5][6] 인간에게 그것은 두 개의 사이클로옥시제 중 하나이다.

역사

Cyclooxygenase(COX)는 아라키돈산에서 프로스타글란딘으로 가는 생합성 경로의 중심 효소다. 이 단백질은 40여 년 전에 격리되어 1988년에 복제되었다.[7][8]

진과 이소자메스

구별되는 유전자 생성물에 의해 인코딩된 COX에는 두 개의 등소성분(Isozymes)이 있는데, COX-1 구성성분(이 효소)과 유도성 COX-2로 표현과 조직 분포에 대한 규정이 다르다. 이 두 대본의 표현은 관련 사이토카인성장요인에 의해 차별적으로 규제된다.[9] 이 유전자는 내피세포에서 혈관신생을 조절하는 COX-1을 암호화한다. 또한 COX-1은 세포 신호 전달과 조직 자기장 유지에도 관여한다. 함수

시클로옥시제네아제(COX)로도 알려진 프로스타글란딘-엔도페록세아제(PTGS)는 프로스타글란딘 생합성의 핵심 효소다. 인산화효소 A2의 효소 활성에 의해 sn-2 에스테르 결합 부위의 막인인산화물에서 방출되는 자유 아라키돈산을 프로스타글란딘(PG) H2로 변환한다. 반응에는 사이클록시제네아제(dioxygenase)와 하이드로페록시디아제(peroxidase) 활동이 모두 포함된다. 사이클록시제네아제 활성은 두 개의 산소 분자를 아라키돈산이나 리놀레산아이코사펜타에노산과 같은 대체 폴리불포화지방산 기질에 통합한다. 아라키돈산의 신진대사는 효소의 수산화효소 활성에 의해 해당 알코올인 PGH2로 감소하는 중과산화질소인 PG2를 형성한다.

임상적 유의성

COX-1은 아스피린과 같은 비스테로이드성 소염제(NSAID)에 의해 억제된다. 혈소판 COX-1의 주요 제품인 트롬박산A2는 혈소판 집적을 유도한다.[20][21] COX-1의 억제는 저선량 아스피린이 심장 질환을 줄이는데 효과적인 이유를 설명하기에 충분하다.
