콜라겐 알파-1(COL4A1) 체인(COL4A1)은 인간에서 13번 염색체의 COL4A1 유전자에 의해 인코딩되는 단백질이다.[5][6] 그것은 보편적으로 많은 조직과 세포 유형으로 표현된다.[7] COL4A1은 IV형 콜라겐의 하위 단위로서 혈관신생에 역할을 한다.[8] 유전자의 돌연변이는 뇌, 근육, 신장, 눈, 그리고 심혈관 계통의 질병과 연관되어 있다.[9][10][11][12] COL4A1 유전자는 또한 관상동맥 질환의 위험 증가와 관련된 27개의 SNP 중 하나를 포함한다.[13]
구조
유전자
COL4A1 유전자는 13q34 대역의 13번 염색체에 존재하며 54개의 exon을 포함하고 있다.[5] 이 유전자는 대체 스플라이싱을 통해 2개의 이소성형을 생성한다.[14]
단백질
COL4A1은 타입 IV 콜라겐 계열에 속하며 짧은 N단자 도메인, 그 중심에 긴 삼중헬리컬 7S 도메인, 그리고 그 C단자에는 비콜라겐 1(NC1) 도메인의 세 가지 도메인을 포함한다.[14][15] 트리플헬리컬 도메인은 중단되는 G-X-Y 반복을 포함하고 있어 도메인의 유연성이 의심된다.[14] NC1 영역은 2개의 트리머릭 캡으로 구성되어 있으며, 각각 2개의 알파 1 조각과 1개의 알파 2 조각이 들어 있으며, 축 터널 주위에 배열된 6배의 프로펠러를 형성하고 있다. 이 두 캡 사이의 상호작용은 큰 평면 인터페이스를 따라 발생하며, 두 캡을 가로지르는 알파 1과 알파 2 체인의 공칭 교차 연계에 의해 안정화된다.[15]
함수
IV형 콜라겐은 지하막의 주요 구조 성분으로 COL4A1 단백질이 2~3개 들어 있다.[9] 따라서 COL4A1은 풍부하며 모든 종류의 지하막에서 발견된다.[10] COL4A1의 NC1 영역은 새로운 모세혈관의 형성을 제어하는 중요한 항혈관신생 분자다.[16] NC1은 α1β1 통합체에 결합하여 혈관 상피세포의 특정 통합 신호 경로를 억제한다. 또한 MAPK 신호 캐스케이드를 억제하여 HIF-1α와 VEGF 표현을 조절한다. 이러한 발견은 NC1의 반원리 기능을 설명할 수 있다.[17]
임상적 유의성
COL4A1 24와 25의 돌연변이는 HANAC(신혈증, 동맥류, 근육경련 등의 자가 지배적 유전성 혈관질환)와 관련이 있다.[18] COL4A1 유전자의 돌연변이가 일부 포렌체팔리, 정신분열증 환자에게서 발생한다는 사실도 확인됐다.[19][20]
인간에게서 콜라겐 타입 IV에 대한 COL4A1 유전자 코딩의 새로운 돌연변이가 중국 가정의 자가 우성 선천성 백내장과 관련이 있는 것으로 밝혀졌다. 이 돌연변이는 영향을 받지 않은 가족 구성원이나 200개의 관련 없는 대조군에서는 발견되지 않았다. 본 연구에서 시퀀스 분석 결과 글리782 아미노산 잔류물이 보존도가 높은 것으로 확인되었다.[11] COL4A1 유전자의 새로운 돌연변이에 대한 이 보고서는 수정체의 생리적 및 광학적 특성에서 콜라겐 타입 IV의 중요한 역할을 강조하는 비신성 자가 우성 선천성 백내장의 첫 보고다.[11]
또한 심혈관 분야에서는 13q34 염색체의 COL4A1과 COL4A2 부위가 관상동맥 질환의 고도로 복제된 위치다. 정상적인 동맥의 벽에서 콜라겐 타입 IV는 부드러운 근육 세포 증식을 억제하는 작용을 한다. 이에 따라 인간 혈관 매끄러운 근육세포에서 콜라겐 타입 IV의 단백질 발현이 SMAD3 단백질과 mRNA의 TGFβ 매개 자극에 의해 조절된다는 것이 입증되었다.[12] 전체적으로 관상동맥질환의 병원성 발생은 COL4A1과 COL4A2 유전자에 의해 조절될 수 있다는 결론이 내려졌다.[12]
이 유전자의 돌연변이와 관련된 자가 열성 뇌병증도 보고되었다.[21]
임상 마커
COL4A1 유전자를 포함한 27 loci의 조합을 기반으로 한 다중 로커스 유전적 위험 점수 연구는 사고 및 재발 관상동맥 질환 사건 모두에 대해 증가된 위험과 스타틴 치료의 임상적 유익성을 확인했다. 연구는 지역사회 코호트 연구(말모 다이어트 및 암 연구)와 1차 예방 코호트(JUPitter 및 ASCOT)와 2차 예방 코호트(CARE 및 PROPIT-TIMI 22)의 4개의 추가 무작위 제어 시험(Randomized controlled trial)을 기반으로 했다.[13]
