Postreplication repair복제 후 복구는 복제 후 발생하는 DNA 손상을 복구하는 것이다.

정확하고 효율적인 DNA 복제는 모든 생물체의 건강과 생존에 중요하다.최적의 조건에서 복제 DNA 중합효소 ,, Δ, α는 모든 세포 주기에서 게놈을 높은 정확도로 효율적으로 복제할 수 있도록 하기 위해 함께 작동할 수 있다.그러나 DNA는 태양 자외선(UV) 방사선과 세포대사의 부산물로 생성된 반응성 산소종(ROS) 등 외생성 및 내생성 유전독성 위협에 끊임없이 도전받는다.손상된 DNA는 복제 중합체의 강직 블록으로 작용할 수 있으며, 따라서 복제 중단 현장에서 불완전한 DNA 복제나 2차 DNA 가닥 파손의 형성을 초래할 수 있다.불완전한 DNA 합성과 DNA 가닥 파단은 둘 다 유전학적 불안정의 잠재적인 원인이다.손상된 다양한 형태의 DNA를 복구하고 유전학적 불안정성을 최소화하기 위해 DNA 수리 메커니즘의 무기가 존재한다.대부분의 DNA 수리 메커니즘은 손상된 가닥을 고정하기 위한 템플릿으로 온전한 DNA 가닥을 필요로 한다.
DNA 손상은 복제 포크로 DNA의 정상적인 효소 합성을 방지한다.[1][2][3][4]게놈의 손상된 부위에서 원핵세포와 진핵세포 모두 다수의 복제 후 복구(PRR) 메커니즘을 활용하여 DNA 복제를 완료한다.화학적으로 변형된 염기는 오류 발생 가능성이[5] 있거나 오류 없는[6] 트랜스전션 중합체 또는 자매 크로마티드와의 유전적 교환을 통해 우회할 수 있다.[7]설탕 인산염 등뼈가 부러진 DNA 복제는 이온화 방사선에 대한 저항을 주는 호몰로 재결합 단백질에 의해 촉진될 가능성이 높다.PRR 효소의 활성은 박테리아 내 SOS 반응에 의해 조절되며 eukaryotes 내에서의 복제 후 체크포인트 반응에 의해 조절될 수 있다.[8][9]
PRR 메커니즘의 해명은 분자생물학 연구의 활성 영역이며, 용어는 현재 유동적이다.예를 들어, PRR은 최근 DNA에 대한 원래의 화학적 수정을 제거하지 않고 복제 후 DNA 손상을 수리하는 경우를 강조하기 위해 "DNA 손상 내성"으로 언급되고 있다.[10]PRR이라는 용어는 손상 베이스 반대편에 있는 단일 변형 후 복원 갭의 수리를 설명하기 위해 가장 자주 사용되었지만, 보다 광범위한 용어가 제안되었다.[8]이 경우 PRR이라는 용어는 복제에 의한 이중 가닥 파손을 복구하는 프로세스를 포함하여 손상된 DNA의 복제를 촉진하는 모든 프로세스를 포괄할 것이다.
흑색종 세포는 자외선에 의한 손상의 일종인 사이클로부탄 피리미딘 다이머 형태의 DNA 손상 후복제술에서 흔히 결함이 있다.[11][12]흑색종 세포에 결함이 있는 것으로 보이는 특별한 수리 과정은 동음이의 재조합 수선이다.[12]사이클로부탄 피리미딘 다이머의 복제 후 수리에 결함이 있으면 흑색종의 주요 원인인 돌연변이가 발생할 수 있다.
