그 분야의 간단한 역사: 생물 전기의 선구자

세포에서 생물 수준에 이르기까지 살아있는 시료에서 전기 치수를 정량적으로 추출하는 골드 스탠다드 기술은 유리 마이크로 전극(또는 마이크로핏), 진동(또는 자기 참조) 전압 프로브 및 진동 이온 선택형 마이크로 전극입니다.전자는 본질적으로 침습적이고 후자는 비침습적이지만, 모두 광범위한 생물학적 모델에서 [52][53][11][54][23]과도한 생리 조건에서 광범위하게 사용되는 초민감하고[51] 반응 속도가 빠른 센서이다.
유리 미세전극은 1940년대에 호지킨과 헉슬리의 거대한 축삭 [55][56]오징어에서의 주요 연구로부터 파생된 흥분성 세포의 활동 가능성을 연구하기 위해 개발되었다.생물학적 검체와 전극을 연결하는 단순한 액체 염분 브릿지로, 맨 전극의 침출성 독소와 산화환원 반응으로부터 조직을 보호합니다.낮은 임피던스, 낮은 접합전위 및 약한 편광으로 인해 은 전극은 전극 [57]표면에서 가역적 산화환원반응을 통해 발생하는 전류에 대한 이온의 표준 변환기이다.

초기 개발에서의 역할

인간의 장애와 수많은 마우스 돌연변이는 생체 전기 신호가 인간의 발달에 중요하다는 것을 보여준다(표 1과 표 2).이러한 영향은 이온 채널을 교란시키는 돌연변이로 인해 발생하는 인간 질환인 채널병증과 광범위하게 연관되어 있다.

전압 게이트2+ Ca 채널인 CaV1.2의 돌연변이는 심각한 심장 부정맥(롱 QT)을 발생시키는 티모시 증후군으로 이어지며, 이는 구개열 또는 고궁도 입천장, 미세구개, 낮은 귀, 신디케이트 [119][120]및 완상동맥을 포함한 Andersen-Tawil 증후군과 같은 신디 및 유사한 두개 안면 결손상과 함께 발생한다.이러한 채널로피스는 드물지만, 기능적인 이온 채널이 발달에 중요하다는 것을 보여줍니다.또한 일부 이온 채널을 대상으로 하는 항간질 약물에 대한 자궁 내 노출은 구강 [121][122][123][124][125]구개열과 같은 선천성 결함의 발생률을 증가시킨다.이온 채널의 유전적 및 외인적 교란 효과는 개발에서 생체 전기 신호의 중요성을 통찰합니다.
상처 치유 및 세포 유도 역할
생체 전기 구배에서 가장 잘 알려진 역할 중 하나는 상처 치유 중에 사용되는 조직 수준의 내인성 전기장이다.상처 관련 전기장은 신경 펄스 및 근육 수축에 비해 약하고 덜 변동하며 즉각적인 생물학적 반응을 보이지 않기 때문에 연구하기가 어렵다.진동 및 유리 마이크로 전극의 개발은 상처가 실제로 측정 가능한 전류와 전기장을 [40][126][59][127][128][129]생성하며, 중요한 것은 지속된다는 것을 증명했습니다.이러한 기술은 각막 및 피부 상처의 상처 전장/전류를 더욱 특성화할 수 있도록 하며, 이는 이러한 전기적 현상에 대한 능동적인 조절을 시사하는 능동적인 공간적 및 시간적 특징을 보여준다.예를 들어, 상처의 전류는 상처 가장자리에 항상 가장 강하며,[130][131][61] 상처는 점차 증가하여 부상 후 약 1시간 후에 최고점에 도달한다.당뇨병 동물의 상처에서는 상처의 전기장이 현저하게 [132]손상된다.창상 전류/장의 생성 및 조절 메커니즘을 이해하면 창상 치유를 위해 전기적인 측면을 조작하는 새로운 접근법이 밝혀질 것으로 예상된다.
상처 부위의 전기장은 어떻게 만들어집니까?상피는 이온을 능동적으로 펌핑하여 차등 분리한다.각막상피에서는 예를+ 들어 Na, K가+ 최루액에서 세포외액으로 안쪽으로 운반되고− Cl이 세포외액에서 최루액으로 운반된다.상피 세포는 단단한 접합부로 연결되어 주요 전기 저항 장벽을 형성하며, 따라서 상피 전체에 걸쳐 전기적 경사(TEP)[133][134]를 형성합니다.모든 상처에서 발생하는 것처럼 상피 장벽을 부수면 상피 시트의 단단한 접합부에 의해 확립된 높은 전기 저항을 뚫는 구멍이 생성되어 상피가 국소적으로 단락됩니다.따라서 TEP는 상처에서 0으로 떨어집니다.단, 정상적인 이온수송은 권선단부(일반적으로 1mm 미만)를 넘어 울리지 않은 상피세포에서 계속되며, 권선단부에서 양의 전하가 흐르고 권선단부에서 음극으로 안정된 횡방향 전계(EF)를 확립합니다.피부에서도 TEP가 발생하며, 피부에 상처가 생기면 상피 장벽 기능이 회복되어 상처의 단락이 종료될 때까지 유사한 상처 전류와 장이 발생한다.이온의 운반을 자극하거나 억제하는 약리작용으로 권상전계를 조작하면 권상전계도 각각 증가 또는 감소한다.상처 치유는 각막 [130][131][135]상처에서 그에 따라 빨라지거나 느려질 수 있다.
