C1QBP
C1QBP보체성분 1Q 서브성분 결합단백질인 미토콘드리아는 C1QBP [5][6][7]유전자에 의해 인체 내에서 암호화되는 단백질이다.
인간 보체 하위 성분 C1q는 혈청 보체 시스템의 첫 번째 성분을 생성하기 위해 C1r 및 C1s와 관련된다.이 유전자에 의해 암호화된 단백질은 C1q 분자의 구상 머리에 결합하고 C1 활성화를 억제하는 것으로 알려져 있다.이 단백질은 또한 히알루론산 결합 [7]단백질과 함께 사전 mRNA 스플라이싱 인자 SF2의 p32 서브유닛으로 확인되었다.
단백질 서브유닛
C1QBP는 길이 282개의 아미노산이며, N말단 73개의 아미노산 잔기를 분해하여 성숙한 C1QBP를 생성한다.C1QBP는 환원 및 비환원 조건 하에서 SDS-PAGE 겔에서 33kDa 내외의 단량체로 나타나지만 크기-제외 크로마토그래피(겔 여과)[8]에서는 트리머로 이동한다.
단백질 구조
C1QBP의 2.25Ω 분해능 결정 구조는 대칭을 나타내는 호모트리머 고리를 보여준다.개별 서브유닛은 비공유 상호 작용에 의해 함께 유지되며, 직경 20Ω의 중심 공동을 가진 도넛 모양의 4차 구조를 형성합니다.C1QBP의 각 서브유닛은 7개의 β스트랜드(β1~β7)와 3개의 α헬리시(α1~α3)를 가진다.C1QBP는 용해면에 음전하를 띠는 반면 막면은 [9]양전하를 띠게 된다.
상호 작용
C1QBP는 단백질인산화효소 D1,[10] BAT2,[11] PRKCD,[10] PKC[10] 알파 및 단백질인산화효소 [10]Mζ과 상호작용하는 것으로 나타났다. C1QBP의 다른 상호작용 파트너로는 박테리아,[12] 바이러스, 플라스모듐 팔시파룸 [14]등의 병원균에서 단백질 도메인이 있다.피브리노겐, FXII 및 HK를 포함한 혈장 단백질은 아연 의존적인 방식으로 C1QBP와 상호작용하는 것으로 입증되었다.[15][16]최근 종양 호밍 펩타이드인 LyP-1(CGNKRTRGC)은 종양 발현 [17]세포에서 C1QBP에 선택적으로 결합하는 것으로 나타났다.
