SOX17SRY박스17은 SOX17 유전자에 의해 인체 내에서 암호화되는 단백질이다.[5]
인간 SOX17 궤적에서의 규정
이 유전자는 염색체 8 q11.23에 위치한 SOX(SRY 관련 HMG 상자) 계열의 전사 인자를 코드한다.유전자 본체가 CTCF 루프 [6][7][8]영역 내에서 분리되었습니다.SOX17의 약 230kb 업스트림에서 SOX17 규제 [9][10]요소로 구성된 조직 특이적 차등(하이포) 메틸화 영역(DMR)이 식별되었습니다.특히 DMR에는 SOX17 [11]궤적에서 가장 원위적인 최종 내배엽 고유의 인핸서가 포함되어 있습니다.SOX17 자체는 최근 소위 토폴로지적으로 절연된 유전자(TIG)로 정의되었습니다.정의당 TIG는 CTCF 루프 도메인 내의 단일 단백질 코딩 유전자(PCG)로, 주로 개발 규제 기관으로 강화되며 3D 루프 도메인 [12]아키텍처를 통해 매우 엄격하게 제어될 것을 제안한다.
개발 중인 기능
SOX17은 척추동물의 배아발달 조절과 내배엽세포 운명 결정에 관여한다.암호화된 단백질은 TGF 베타 시그널링(Activin)과 표준 WNT 시그널링(Wnt3a)[13][14]의 다운스트림으로 작용합니다.특히 각 세포주기(초기 G1상) 내에서 SMAD2/3의 정확한 인산화(인산화)는 기수 내배엽 유전자(예를 들어 SOX17)가 최종 내배엽 [15]계통에 더욱 진입하는 데 중요하다.또한 SOX17의 중심성 CTCF 경계가 초기 확정적 내배엽 분화에 있어 섭동함으로써 현저한 발육부전 및 소위 포획 세포상태와 같은 중배엽이 발생하게 되며, 이는 이소성 SOX17 [16]발현으로 구제할 수 있다.Xenopus gastrulae에서는 SOX17이 Wnt 반응을 수정하는 것으로 나타났으며, 여기서 SOX17과 β-catenin/Tcf 전사 [17]복합체 간의 기능적 상호작용을 통해 Wnt/β-catenin 전사의 게놈 특이성이 결정된다.
