SIRT4 로도 알려진 Sirtuin 4는 사람 에게서 SIRT4 [5] [6] 유전자에 의해 암호화되는 미토콘드리아 단백질 이다.SIRT4는 효모 Sir2 단백질의 상동어 인 포유동물 Sirtuin 계열의 단백질이다. SIRT4는 NAD+의존성 탈아세틸화효소 활성을 나타낸다.
기능. SIRT4는 미토콘드리아 글루탐산탈수소효소 1의 활성을 억제하여 아미노산에 [7] 반응하여 인슐린 분비를 억제 하는 미토콘드리아 ADP-리보실전달효소이다. SIRT4에 의한 말로닐-CoA 탈탄산화효소 탈아세틸화 는 효소 활성을 억제하여 근육 [8] [9] 및 간세포의 지방산 산화를 저해 한다. SIRT4는 간세포의 [9] 지방산 산화를 하향 조절하는 페르옥시좀 증식기 활성화 수용체 알파(PPAR-α)에 억제 효과가 있다. ADP/ ATP 트랜스로카아제2 (ANT2)의 탈아세틸화 는 미토콘드리아 [9] 분리 를 감쇠시킴으로써 세포 ATP를 증가시킨다.
임상적 의의 SIRT4는 미토콘드리아 종양 억제 [9] 단백질이다. SIRT4의 과잉발현 은 글루타민 대사 [9] [10] 저하에 의해 암세포 증식을 억제한다.
