프로사포신
ProsaposinPSAP라고도 알려진 프로사포신은 인간에게 PSAP 유전자에 의해 암호화된 단백질이다.[5]
이 보존도가 높은 당단백질은 사포신 A, B, C, D의 4가지 갈라진 제품의 전구체다.사포신은 스핑골리피드 액티베이터 PrO[S]teINs의 약자다.[6]전구단백질의 각 영역은 약 80개의 아미노산 잔류물이며, 시스테인 잔류물과 글리코실화 부위가 거의 동일하다.사포신 A-D는 주로 라이소솜 구획에 국소화하며, 여기서 그들은 짧은 올리고당류 집단을 가진 글리코스포싱콜리피드의 포타볼리즘을 촉진한다.전구 단백질은 분비성 단백질과 일체성 막 단백질로 둘 다 존재하며 신경영양 활동을 한다.[5]

가족구성원
- 사포신 A는 고립되기 전에 프로사포신 cDNA에서 N단자 도메인으로 식별되었다.4-메틸룸벨리페릴-β-글루코사이드, 글루코세레브로사이드, 갈락토세레브로사이드의 효소 가수분해를 자극하는 것으로 알려져 있다.[8]
- 사포신 B는 최초로 발견되었으며, 아릴술파타아제 A에 의한 황산화물의 가수 분해에 열안정 인자로 필요한 것으로 밝혀졌다.그것은 스핑골리피드 활성제 단백질-1(SAP-1), 황산염 활성제 단백질, GM1 강리오사이드 활성제, 디스페신, 비특수 등 많은 다른 이름으로 알려져 있다.[9]이 특별한 사포신은 효소 자체가 아닌 기판과의 상호작용을 통해 많은 효소를 활성화시킨다는 것이 관찰되었다.
- 사포신 C는 글리코실세라마이드에 의한 글리코세레브로이드와 갈락토세레브로이드에 의한 글리코세레브로이드의 가수분해를 자극하는 두 번째 사포신이었다.
- 사포신 D는 현시점에서는 조사부족으로 잘 알려져 있지 않다.프로사포신의 cDNA 순서에서 사포신A와 같이 예측되었다.효소 자극은 이 특정한 당단백질에게 매우 구체적이며 완전히 이해되지는 않는다.[7]
- GM2A(GM2 ganglioside activator)는 SAP 계열의 멤버로 간주되어 SAP-3(spingolipid activator 단백질 3)로 불렸다.[10]
구조
모든 사포신은 약 80개의 아미노산 잔류물을 함유하고 있으며 6개의 균등하게 배치된 시스테인, 2개의 프로라인, 글리코실화 부위(사포신 A에 2개, 사포신 B, C, D에 각각 1개씩)가 있다.[7]사포신 특성은 극도의 열안정성, 이황화 연계의 풍부함, 그리고 대부분의 보호제에 대한 저항성이기 때문에 극히 콤팩트하고 강직하게 이황화 연계된 분자로 가정한다.각 사포신은 자극에 중요한 α-헬리컬 구조를 가지고 있는데, 이 구조는 4.5의 pH에서 최대치이기 때문에 많은 라이소솜 하이드롤에 최적이다.[7]이 나선 구조는 모두(특히 첫 번째 부위에서) 볼 수 있지만, 사포신은 N-end의 첫 24개의 아미노산으로 인해 β-시트 구성이 있을 것으로 예측되어 왔다.[9]
함수
그들은 아마도 막 주위로부터 지질 기질을 분리하여 용해성 분해 효소에 더 쉽게 접근할 수 있게 함으로써 작용한다.여기에는 4개의 Saposin-B 도메인이 포함되며, 단백질 분해 후 활성 사포신과 활성화 반응에서 제거된 2개의 Saposin-A 도메인이 포함된다.사포신-B 영역은 다른 단백질에서도 발생하는데, 이들 중 다수가 세포막의 용해에서 활동한다.[14][15]
