프로프로테인 변환효소 서브틸리신/케신 타입 6은 인간에서 15번 염색체에 위치한 PCSK6 유전자에 의해 인코딩되는 프로프로테아제다.[5][6]Pcsk6는 칼슘에 의존하는 세린내포효소로, 전구 단백질의 변환 후 수정을 '대기' 형태에서 분해된 '활성' 형태로 촉진한다.[5]액티브 Pcsk6는 성장 인자 변환 β,[7] 프로 알부민,[8] 폰 윌레브란트 인자,[9] 코린 등의 기판을 처리하는 것으로 보고되었다.[10]임상적으로 Pcsk6는 좌우 비대칭,[11] 뇌의 구조적 비대칭,[12] 손동작,[13][14][15] 종양 진행, [16]지혈,[9][8][7] 심혈관 질환 등의 역할을 하는 것이 제안되고 있다.[10][17]
이 유전자가 인코딩한 단백질은 아빌리신 같은 프로프로테아제 변환효소 계열에 속한다.이 가족의 구성원들은 잠재된 전구단백질을 생물학적으로 활동적인 생산물로 처리하는 프로프로테아 변환물이다.이 인코딩된 단백질은 칼슘 의존성 세린 내복단백질로, 쌍을 이룬 기본 아미노산 처리장에서 전구단백질을 분쇄할 수 있다.기질로는 변형성장인자 베타 관련 단백질,[7] 프로알부민,[8] 폰 윌레브란드 인자,[9] 코린 등이 있다.[10]대안으로 서로 다른 ISO 양식을 인코딩하는 분할된 대본 변형이 식별되었다.[6]
임상적 유의성
개발 중:개발 전반에 걸쳐 pcsk6의 공간적, 시간적 표현은 BMP와 노달을 포함하는 TGFβ 관련 분화계수를 활성화하여 발생생성을 조절한다.[7][18]Pcsk6의 높은 수준은 삽입 지점과 외측부 세포에서 검출되었다.[19]노달과 BMP의 적절한 구배 규제는 미식,[20] 근위축,[21] 좌우축 패터닝의 확립에 매우 중요하다.[22]
개발 Pcsk6 Kockout 연구 결과 Pcsk6가 부족한 생쥐 배아는 노달과 BMP 신호의 조절이 잘못되어 전축과 우축의 사양에 차질이 생겨 이형성, 좌폐 이형성, 그리고/또는 두개골 기형이 발생하는 것으로 밝혀졌다.[11]
인간에서 Pcsk6 VNTR 다형성은 전두엽과 측두엽의 구조적 비대칭성,[12] 그리고 손의 정도와 관련이 있다.[13][14]
심혈관 질환:pcsk6는 심혈관 질환의 지표와 요인으로 관심이 높아지고 있다.Pcsk6 KO 생쥐는 혈압의 심방 내유성 펩타이드 조절에 필수적인 프로코린 활성화의 실패로 소금에 민감한 고혈압이 발병하는 것으로 나타났다.[10]고혈압 환자는 PCSK6 유전자에 G/A 돌연변이가 있어 Pcsk6 촉매영역에서 Asp282AsN(D282N) 대체가 일어나 코린 처리를 방해하는 것으로 밝혀졌다.[10]혈관 리모델링에서는 Pcsk6가 MMP14를 활성화해 PDGFB에 대응한 원활한 근육세포이동을 유도하는 것으로 나타났다.[17]Pcsk6가 녹아웃되었을 때, 생체내 경동맥 경동맥 레깅에 대한 근거리 과대증 반응이 낮아졌다.[17]
