가능성이 있는 헬리코아제 세나탁신은 인간에게 SETX 유전자에 의해 암호화된 효소다.[5][6][7]
이 유전자는 302kDa 단백질인[8] 세나탁신이라는 단백질을 암호화한다.
순서 및 구조
인간 SETX와 효모 Sen1 사이에는 높은 동음이의어가 있다.효모 속의 Sen1은 RNA/DNA 헬리코아제로서 특히 헬리코아제 영역에서 이들 유전자 사이의 보존성이 높은 시퀀스는 인간의 SETX가 유전자 발현과 게놈 안정성 유지에 있어 유사한 역할을 할 수 있음을 나타낸다.Sen1에서 N-terminus는 RNA 중합효소 II, 리보누클리스 III, NER 인자 Rad2/XPG의 C-단자 영역과의 상호작용을 보였다.한편, C-terminus는 DNA/RNA 헬리코아제 활동을 암호화한다.[9]마찬가지로 SETX는 다른 단백질과의 상호작용에 관여할 가능성이 높은 N단자를 가진 세나탁신 단백질을 암호화한다.세나탁신은 RNA 중합효소 II와 폴리(A) 결합 단백질과 상호작용한다.C-단자에서는 세나탁신(senataxin)이 DEAD 박스 헬리코아제 영역을 가지고 있다.[10]
함수
체내 많은 조직에서 세나탁신이 널리 표현되지만, 세나탁신의 세포 역할은 완전히 이해되지는 않는다.그러나, 현재 SETX의 연구와 조사 호몰로그램에 근거해, 세나탁신은 DNA 손상 대응(DDR)의 필수적인 요소로서, R-루프 해결, 전사 종료, 게놈 안정성 유지에 중요한 역할을 하는 것으로 생각된다.[11]
SETX는 DNA 손상 반응에서 다른 단백질과 함께 작업해 DNA 손상 복구와 게놈 안정성 유지에 관여하는 것으로 의심받고 있다.R 루프는 특정 로키에서 전사 및 복제가 동시에 발생하는 경우와 같이 복제 스트레스로 인해 발생할 수 있다.이것은 긴 유전자를 기록할 때 종종 발생한다. 왜냐하면 그 유전자의 전사에는 한 번의 복제보다 더 오랜 시간이 걸릴 수 있기 때문이다.복사와 전사 기계가 충돌하면 R 루프가 형성되고 이중으로 좌초된 균열이 형성될 수 있다.[12]이들 충돌 현장에서는 SETX가 DNA 손상 표식인 53BP1과 공동 국산화되는 것으로 나타났다.[13]또한, SETX는 균질 재조합 수리를 촉진하고 번역을 방지하기 위해 관찰되었다.[14]DNA 손상 수리에 있어 SETX의 역할을 더욱 지원하기 위해, SETX는 다른 많은 DDR 요소들과 공동 국산화한다.예를 들어, BRCA1은 R-루프 구조의 일부인 취약한 단일 좌초 DNA로 인해 발생하는 DNA 변이를 방지하는 R-루프를 제거하기 위해 SETX를 모집하는 것도 보였다.[15]SETX는 RAD51 로딩에 관여하여 이중 Strand Break 수리에 관여할 수 있으며, 이는 동질 재조합을 통한 이중 Strand Break 수리에 있어 중요한 단백질이다.[16]

임상적 유의성
SETX는 오쿨로모터 아프락시아 2형(AOA2)과 소아형 근위축성 측경화증(ALS4)에서 변이된 것으로 확인됐다.[18]ALS4 세포에서 SETX는 헬리코아제 기능을 더 많이 갖도록 변이되어 R-루프 수치가 평상시보다 낮아져 이상 TGF-β 신호를 유발하고 뉴런 사망을 일으킨다.[19]AOA2 세포는 세나탁신 기능 상실과 비정상적으로 높은 R루프 수준을 보여준다.[20]AOA2나 ALS4와 같은 신경질환은 단백질 골재가 비정상적으로 축적되는 경우가 많으며, 연구결과 SETX가 단백질 골재를 맑게 하는 데 관여하는 유전자를 조절함으로써 자가포화에 필수적인 역할을 할 수 있다는 것이 밝혀졌다.[21]
