플로싱
Floxing유전학에서 플로싱은 두 개의 LOX P 부위 사이에 (그 후 플로싱된 것으로 알려져 있는) DNA 서열을 샌드위치하는 것을 말한다.이 용어는 "LoxP에 의한 플랭킹/플랭킹"이라는 문구에 기초하여 구성된다.LoxP 사이트 간의 재결합은 Cre 재조합법에 의해 촉진된다.유전자를 플록스(flox)하면 Cre-Lox 재조합이라는 과정에서 삭제(knocked out), 번역 또는 반전시킬 수 있다.[1] 유전자의 플러싱은 연구자들이 유전자 발현의 공간적, 시간적 변화를 가질 수 있게 해주기 때문에 과학적인 모델 시스템의 개발에 필수적이다.[2]게다가 쥐와 같은 동물들은 인간의 질병을 연구하는 모델로 이용될 수 있다.따라서 크레록스 시스템은 인간의 질병과 약물 개발을 연구하기 위해 쥐에 유전자 발현을 조작하는 데 사용될 수 있다.[3]예를 들어, 크레록스 시스템을 사용하여, 연구원들은 생쥐 모델에서 암의 발생과 진행에 대한 그들의 역할과 종양 억제기 유전자를 연구할 수 있다.[4]
연구에 사용
유전자를 플러싱하면 삭제([5][6]노킹 아웃), 번역 또는 삽입[7](Cre-Lox 재조합의 다양한 메커니즘을 통해)할 수 있다.
유전자의 플러싱은 유전자 발현의 공간적, 시간적 변화를 가능하게 하기 때문에 과학적인 모델 시스템의 개발에 필수적이다.비전문가의 관점에서, 이 유전자는 과학자가 선택한 언제든지 생체내 특정 조직에서 녹아웃(비활성화)될 수 있다.그러면 과학자들은 녹아웃된 유전자의 영향을 평가하고 유전자의 정상적인[8] 기능을 확인할 수 있다.이는 수정에서 출발하는 유전자가 없는 것과 다른데, 유기체의 발전에 필수적인 유전자의 불활성화 또는 상실로 세포의 정상적인 기능을 방해하고 생존 가능한 자손의 생성을 방해할 수 있다.[9]
삭제 메커니즘
삭제 이벤트는 유전자 편집 실험을 수행하는 데 유용하다.삭제하려면 동일한 방향을 향하는 loxP 사이트로 관심 세그먼트를 플러싱해야 한다.Cre 재조합효소는 단방향 loxP 부지를 검출하고 DNA의 플록스 부분을 제거한다.[10]성공적으로 편집된 클론은 동일한 Cre-loxP 시스템을 사용하여 제거할 수 있는 선택 마커를 사용하여 선택할 수 있다.[10]동일한 메커니즘을 사용하여 동일한 메커니즘을 수행하지만 다른 효소를 갖는 FRT/Flp 사이트를 도입함으로써 조건부 알레르기를 만들 수 있다.
반전 메커니즘
반전 현상은 유전 물질의 양을 유지하는 데 유용하다.반전 유전자는 비정상적인 표현형과 자주 연관되지 않는데, 이는 반전 유전자가 일반적으로 생존할 수 있다는 것을 의미한다.[11]삽입을 초래하는 크레-록스P 재조합은 관심 유전자를 플록스하기 위해 loxP 사이트가 서로 지향하는 것을 필요로 한다.크레 재조합을 거치면 roxP 사이트에 의해 플러싱된 지역이 반전되는데,[12] 이 과정은 영구적이지 않고 되돌릴 수 있다.[13]
번역 메커니즘
번역작용은 loxP가 두 개의 서로 다른 DNA 분자에 단방향으로 유전자를 플록스할 때 일어난다.크레 재조합효소는 그 후 두 DNA 분자 사이에 번역을 생성하기 위해 사용되며, 한 DNA 분자에서 다른 DNA 분자로 유전 물질을 교환하여 플로크된 두 유전자의 동시 번역을 형성한다.[14][15]
연구에서의 일반적인 애플리케이션
심장근육세포(심장근육 조직)는 심장근육세포에 매우 특이하고 연구자들이 고효율 재조합을 수행하는 데 사용할 수 있는 크레 재조합제의 한 유형을 나타내는 것으로 나타났다.이는 - myosin 헤비체인 프로모터( -MyHC)에 의해 표현되는 크레의 유형을 사용하여 달성된다.이러한 재조합은 체내 심장 조직에만 특정한 방식으로 유전자를 교란시킬 수 있으며, 대부분 조정기로 사용하기 위해 심장의 조건부 녹아웃을 만들 수 있다.[16]
예를 들어 재조합효소를 α {\\alpha } -MyHC 프로모터와 함께 사용하면 심장에서만 플록스 유전자가 비활성화된다.또한 이러한 녹아웃은 유도할 수 있다.몇몇 마우스 연구에서는 타목시펜을 사용하여 크레 재조합을 유도한다.[17][18]이 경우 크레 재조합효소는 리간드 결합 영역(LBD) 내에 돌연변이를 포함하는 생쥐 에스트로겐 수용체(ER)의 일부에 융합된다.돌연변이는 수용체를 비활성 상태로 만들어 열충격단백질 70, 90(Hsp70, Hsp90)과 같은 세포단백질과의 상호작용을 통해 부정확한 국산화(localization)로 이어진다.타목시펜은 Cre-ER에 결합하고, Cre-ER 융합 단백질이 핵으로 들어가 플로렉스 유전자에 재조합을 할 수 있도록 하는 샤페론과의 상호작용을 방해한다.[19][20]또한 크레 재조합효소는 특정 열충격 요소(HSE)의 제어 하에 있을 때 열에 의해 유도될 수 있다.[21][22]
